Microphthalmia Associated Transcription Factor (MITF) is a transcription factor that plays a key role in cell differentiation, development and maintenance of physiological functions. It belongs to the MiT (microphthalmia transcription factor) basic helix-loop-helix family. Microphthalmia Associated Transcription Factor (MITF) has at least MITF-A, MITF-B, MITF-C, MITF-H and MITF-M subtypes, each of which has a different structure due to its unique N-terminal coding exon. MITF has many interacting targets, such as β-catenin, which can change β-catenin transcriptional activity and thus affect gene expression; GLI2, TGF-β, etc., which affect cell processes by inhibiting MITF transcription. In addition, miR-204/211 is also an important target regulated by MITF and participates in the process of cell differentiation. MITF is essential for the development, maintenance and differentiation of melanocytes, regulating the transcription of melanin production enzyme genes; in retinal pigment epithelial cells, it promotes cell differentiation by regulating miR-204/211 expression. MITF is associated with a variety of diseases, and its p.E318K mutation is associated with an increased risk of melanoma and renal cell carcinoma. In renal cell carcinoma, patients carrying this mutation often present with bilateral multifocal type 1 papillary renal cell carcinoma; MITF gene abnormalities are also associated with Waardenburg syndrome, Tietz syndrome and other auditory-pigmentary diseases[1][2][3][4][5][6][7][8][9].
